Overview

The Morris Water Maze (MWM) is a classic assay of spatial learning and memory in rodents. Established by Richard Morris in 1981, its core design places a hidden escape platform in a circular pool. How animals find and climb onto the platform reflects spatial cognition and memory.

MWM is widely used to study hippocampal spatial learning mechanisms, memory deficits in Alzheimer’s and other cognitive models, entorhinal and related circuits, aging or brain injury effects on cognition, and cognitive-enhancing drug discovery and efficacy evaluation.

Methods

  1. 1Acquisition (place navigation): Train mice 4 trials/day for 7 days, ~1 h apart. Release from N/S/W/E facing the wall; record latency to find the submerged platform within 60 s. If not found, guide the mouse to the platform and leave it there for 30 s.
  2. 2Probe trial (day 8): Remove the platform and release from the opposite quadrant. Record latency to the former platform location, platform crossings, path in the platform annulus, and time in the target quadrant over 60 s.
